海豚组织中多氯联苯(PCBs)气相色谱测定的改良方法研究

An ameliorated method for determination of polychlorinated biphenyls(PCBs) in tissues of dolphin by gas chromatography

  • 摘要: 研究和建立了一种测定海豚体内不同组织中多氯联苯(PCBs)前处理的改良方法,即将海豚组织分为低脂样品和高脂样品,分别经萃取、净化、浓缩等处理后,用Agilent 6890N型气相色谱仪、63Ni放射源ECD检测器测定,采用保留时间定性和峰面积外标法定量。结果显示,加标回收率为75.8%~119.2%,精密度为5.6%~11.2%,方法检出限为0.126~0.642 ngg-1,说明该方法的准确性、可重复性符合海洋生物体痕量有机污染物分析要求。海豚组织样品的实际测定结果表明,该方法适合海豚体内PCBs含量的分析测定。

     

    Abstract: An ameliorated pretreatment method for determination of polychlorinated biphenyls(PCBs)in tissues of dolphin has been established. After pretreated in turn by the extraction, purifying for the high-fat tissues and low-fat tissues of dolphin samples, the PCBs were determined by a gas chromatography (Agilent 6890N) equipped with an ECD (electron capture detector). Identification and quantification of PCBs were performed by retention time and external standard method respectively. The results showed that the recovery was between 75.8% and 119.2%, the coefficient of variation was 5.6%~11.2%, and the detection limits of the method were 0.126~0.642 ngg-1. The data of accuracy and repeatability are satisfied for analysis on the trace amounts of organic contaminants in marine organisms. The results of practical analysis for dolphin samples showed that the method was suitable to determine the contents of the PCBs in the dolphins.
