罗非鱼体内氟甲砜霉素的高效液相色谱测定方法研究

High-performance liquid chromatographic determination of florfenicol in tilapia(Oreochromis niloticusO.aureus)

  • 摘要: 建立了罗非鱼(Oreochromis niloticusO.aureus)血浆、肌肉和肝脏内氟甲砜霉素的高效液相色谱测定方法(HPLC法)。采用内标法和峰高法定量,方法准确,重复性好,灵敏度较高。8、1和0.125 gmL-1 3个浓度水平的加标回收率为89.16% ~96.13%;对应的批内精密度和批间精密度分别为2.21% ~6.55%和2.11% ~7.71%。方法检出限为0.03gmL-1。结果表明,此方法非常适合用于罗非鱼体内氟甲砜霉素的动力学和残留研究。

     

    Abstract: A liquid chromatographic (HPLC) method for determination of florfenicol in the plasma, muscle and liver of tilapia (Oreochromis niloticus O.aureus) was developed. For quantitation, the peak-height measurements and the internal standard were used. The study showed that the linearity and repeatability was good. The method was validated with blank samples fortified at 8, 1 and 0.125 gmL-1, the mean recovery was between 89.16% and 96.13%, the coefficient of variation was lower than 7.7%, and the detection limit for flofenicol was 0.03 gmL-1. All the results indicate its usefulness for conducting pharmacokinetics and residues studies on tilapia.
