长臀鮠AFLP反应体系的建立

The establishment of AFLP analysis system in research of armorhead catfish, Cranoglanis bouderius

  • 摘要: 以珠江长臀鮠(Cranoglanis bouderius)为试验材料,建立了长臀鮠扩增酶切片段长度多态性(AFLP)分析体系,对其分析过程中的DNA提取、双酶切、单酶切、连接、预扩增、选择性扩增、 银染等步骤进行了检测, 并对反应条件进行了优化。用 Pst I/Taq I、EcoR I/Mse I 等2组双酶切组合构建长臀鮠的AFLP指纹图谱,条带清晰可辨,扩增信号强,得到清晰的长臀鮠AFLP指纹图谱。

     

    Abstract: AFLP (amplified fragment length polymorphism) analysis system was established in research of Pearl River armorhead catfish, Cranoglanis bouderius. The several steps for AFLP analysis were examined, including DNA extraction, double-restriction endonuclease reaction, single-restriction endonuclease reaction, adadpter ligation reaction, preamplification reaction,selective amplification reaction and Ag-NORs reaction. The AFLP fingerprinting experiments of C. bouderius with primer combination, E+AAG/M+CTG and P+ATT/T+ACA, were performed, which had about 50 clear and bright amplification bands.
