豹纹鳃棘鲈尾部溃烂症病原菌的鉴定与药敏试验

Identification and drug sensitive test of bacterial Pathogens from Plectropomus leopardus with tail fester disease

  • 摘要: 2011年海南会文烟堆某豹纹鳃棘鲈(Plectropomus leopardus)养殖场突发大量死亡,病鱼体色发白,尾鳍溃烂。从病鱼病灶处分离出1株菌X11YD05,回归感染证实为患病豹纹鳃棘鲈病原菌,对平均体质量为30.8 g的斜带石斑鱼(Epinephelus coioides)的半数致死量(LD50)为1.67105 cfug-1。为进一步确定病原菌的分类学特征,扩增其16S rDNA、toxR、rpoD、mreB、topA和rctB等6种管家基因,构建系统发育树。结果发现,X11YD05与哈维弧菌(Vibrio harveyi)有较高同源率。结合生理生化试验,确定X11YD05为哈维弧菌。16种药敏试验表明该菌仅对氧氟沙星、诺氟沙星、环丙沙星、恩诺沙星、复方新诺明等5种抗生素敏感。

     

    Abstract: In 2011, there was an outbreak in a Plectropomus leopardus farm in Wenhui Town, Yandui Village, Hainan Province. The color of sick fish turned white and the tail fin festered. We isolated a Strain X11YD05 from sick P.leopardus which was proved to be pathogen by artificial infection and the LD50 was 1.67105 cfug-1 in Epinephelus coioides using Karber method. To determine the molecular characteristics of X11YD05, we amplified the16S rDNA, transmembrane regulatory protein (toxR), RNA polymerase 70 factor (rpoD), actin-like cytoskeleton protein (mreB), topoisomerase (topA) and replication origin-binding protein (rctB) by PCR. Sequence analysis indicates that X11YD05 belonged to V.harveyi. Drug sensitivity test shows that X11YD05 was sensitive to norfloxacin, ofloxacin, ciprofloxacin, enrofloxacin and cotrimoxazole.
