黄河鲤对饲料蛋白需求量研究

Research on the feed protein requirement of Cyprinus carpio haematopterus

  • 摘要: 黄河鲤 (Cyprinus carpio haematopterus) 是中国黄河流域长期自然形成的特有重要淡水经济鱼类,其对饲料蛋白的需求量较其他鲤要低。为从生长性状、免疫、代谢等方面探究黄河鲤对不同饲料蛋白含量的适应和利用能力,对初始体质量为 (360.22±1.02) g的黄河鲤投喂蛋白质量分数为22.63%、25.32%、28.15%、31.43%、34.18%的实验饲料。结果表明:蛋白质量分数28.15%组生长性状的体质量增长率、特定生长率和蛋白质效率极显著高于其他组 (P<0.01),而饵料系数显著低于其他组 (P<0.05)。蛋白质量分数28.15%组的生长基因GHIGF-1和蛋白合成基因TOR4EBP2的相对表达量显著高于蛋白质量分数22.63%、31.43%和34.18%组 (P<0.05)。蛋白质量分数28.15%组血清生化中的谷草转氨酶和谷丙转氨酶含量显著低于蛋白质量分数22.63%和34.18%组,但碱性磷酸酶含量显著高于这两组 (P<0.05)。蛋白含量28.15%组免疫酶活中的α-淀粉酶、脂肪酶、胰蛋白酶、超氧化物歧化酶、过氧化氢酶活性及还原性谷胱甘肽含量显著高于蛋白质量分数22.63%和34.18%组 (P<0.05),而丙二醛含量极显著低于其他组 (P<0.01)。蛋白含量34.18%组氨代谢基因RhagRhbgRhcg1的相对表达量显著高于其他组 (P<0.05)。拟合回归分析表明,黄河鲤的最佳饲料蛋白需求量约为28.57%~29.04%。该研究有助于提高饲料蛋白利用率,减少饲料成本,为低氮饲料的研究与开发奠定理论基础,也为优化鲤配合饲料营养配比提供理论依据。

     

    Abstract: Abstarct: The Yellow River carp (Cyprinus carpio haematopterus) is a unique and important freshwater economic fish that has naturally formed in the Yellow River Basin of China for a long time. Its demand for feed protein is lower than that of other carp. In order to explore the adaptation and utilization ability of Yellow River carp to different dietary protein contents in terms of growth traits, immunity and metabolism, yellow carp with initial body weight of (360.22±1.02) g were fed with test diets with protein contents of 22.63%, 25.32%, 28.15%, 31.43% and 34.18%. The results show that the weight gain rate, specific growth rate and protein efficiency in growth traits are highly significantly higher at 28.15% protein content than the other groups(P<0.01), and the feed coefficient is significantly lower at 28.15% than the other groups(P<0.05). Relative expression of growth genes GH, IGF-1 and protein synthesis genes TOR, 4EBP2 are significantly higher at 28.15% of protein content than 22.63%, 31.43% and 34.18% of test groups(P<0.05). Serum biochemistry levels of glutamic and glutamic aminotransferase are significantly lower in protein content 28.15% than in 22.63% and 34.18% test groups, but alkaline phosphatase levels are significantly higher in 22.63% and 34.18% test groups(P<0.05). The enzyme activities of α-Amylase, lipase, trypsin, superoxide dismutase, catalase, etc. And the content of reduced glutathione in the protein content 28.15% test group are significantly higher than those in the 22.63% and 34.18% protein content test groups(P<0.05), and malondialdehyde is highly significantly lower in 28.15% than in the other groups(P<0.01). The relative expression of Rhag, Rhbg and Rhcg1 among ammonia metabolism genes are significantly higher in 34.18% of protein content than other groups(P<0.05). The fit regression analysis show that the optimal dietary protein requirement of yellow river carp is about 28.57%~29.04%. This study helps to improve the feed protein utilization rate, reduce the feed cost, lay a theoretical foundation for the research and development of low nitrogen feed, and provide a theoretical basis for optimizing the nutritional ratio of carp formula feed.
