液熏罗非鱼片在25℃贮藏过程中生物胺的变化

Changes of biogenic amines in liquid-smoked tilapia stored at 25 ℃

  • 摘要: 为考察液熏罗非鱼片在贮藏过程中生物胺的质量分数及其变化, 鱼片采用真空和普通2种包装方式在25℃下贮藏, 使用HPLC-紫外检测器分析腐胺(Put)、色胺(Try)、组胺(His)、苯乙胺(Phe)和亚精胺(Spd)5种生物胺的变化。结果表明, 5种生物胺在30min内得到有效的分离, 并且在0.25~10.0μg.mL-1质量浓度范围内呈良好的线性关系(R2 > 0.999), 最低检测限0.02μg.mL-1, 回收率73.8%~113.6%, 相对标准偏差(RSD)低于10%。在真空包装和普通包装样品中, Put的最终质量分数为(13.94±0.66)和(16.25±0.16)mg·kg-1, His为(8.76±0.17)和(6.38±0.21)mg·kg-1, Spd为(0.85±0.17)和(0.95±0.21)mg.kg-1, 而Try和Phe在贮藏全程未被检出。真空包装能明显延缓且抑制Put质量分数的增加, 但对His和Spd无影响, 且在2种包装的鱼片中检出的His均未超过国家标准。

     

    Abstract: In order to study the changes of biogenic amines(BA)in liquid-smoked tilapia slices during storage, the samples were stored in normal and vacuum packages at 25 ℃ to observe the changes of putrescine, tryptamine, histamine, phenethylamine and spermidine by HPLC-UV detector.The results indicate that the five BAs can be effectively separated within 30 min, and have a good linear relation when concentration is in the range of 0.25~10 μg·mL-1(R2 > 0.999);their lowest limit of detection(LOD)is 0.02 μg·mL-1 and the recoveries range from 73.8 % to 113.6 %; the RSDs are less than 10%.In vacuum package and normal package, the final mass fraction of putrescine are(13.94±0.66)and(16.25±0.16)mg·kg-1, respectively; histamine, (8.76±0.17)and(6.38±0.21)mg·kg-1, respectively; spermidine, (0.85±0.17)and(0.95±0.21)mg·kg-1, respectively.However, tryptamine and phenethylamine can not be detected during the storage.Vacuum package can effectively delay and inhibit the increase of putrescine, but does not affect histamine and spermidine.Additionally, the histamine content in both two packages does not exceed the national standard.
