The sequence analysis of mitochondrial cytochrome b gene and molecular phylogeny of Parargyrops edita

  • The sequence of mitochondrial cytochrome b (Cyt b) gene was amplified from Parargyrops edita using PCR technique and it had a length of 1 140 bp. Then the sequence was aligned with the Cyt b gene sequences of Evynnis japonica, Pagrus auratus, P.major, Argyrops spinifer, Dentex tumifrons, Rhabdosargus thorpei, Acanthopagrus schlegelii and Acanthopagrus berda in GenBank. The results demonstrated that there were 407 nucleotide sites substituted (35.7%)P.edita and E.japonica were the most related among the 9 Sparidae fishes. Their genetic divergence was only 1.5%, so they two might belong to the same genus (Evynnis), which was in consistent with that of the traditional morphological taxon. The genetic divergence of P.edita with P.auratus and P.major were 8.3% and 8.4%, respectively, which showed differentiation at the level between strains. Moreover, they assembled one branch in the molecular phylogeny with the most bootstrap value. Whether the Evynnis genus and Pagrus genus belong to the same genus still needs further study.
